CHIMERIC PLAQUE ASSAY-GROWTH IN TISSUE CULTURE

1. MDCK cells were infected with M2HATM and wild type virus at an MOI of 0.001 PFU per cell and 37 degrees celcius.

For the DM2TMCYT mutants, MDCK cells were infected with the mutant at an MOI of 0.01 PFU per cell and incubated at 37 degrees celcius.

2. Yields of virus in the supernatant were counted every 12 hours post infection via plaque assays.

Would you expect to see a difference in either type of mutation?

Yes. Because of the different temperature conditions set
