To test the efficiency of virus generation (replication):

1. They titrated the M2 mutants and wild-type viruses in the culture supernatant of 293 T cells with MDCK canine kidney cells used for tissue experimentation cells

48 hours post transfection.

2. Count number of plaques (plaque assay)

 **This was done for the point mutations and the chimeric mutations

STOP: Why use MDCK cells?

  Answer Because they can be grown experimentally easily and it's cheap
